Outside Out Patch

The outside-out patch is an excised patch-clamp configuration that exposes the extracellular surface of a cell membrane, allowing researchers to study ion channels and membrane receptors in a controlled environment. It forms when a whole-cell patch is gently withdrawn from the cell, causing the membrane to reseal with its outer leaflet facing the surrounding solution while the intracellular face remains connected to the recording pipette. This orientation enables precise application of neurotransmitters, drugs, and other ligands, along with rapid solution exchange, while electrical recordings measure channel currents. Outside-out patches are widely used to investigate channel gating, receptor pharmacology, ion selectivity, and cellular signaling mechanisms.

Neuron cell membranes are populated with ion channels that control the movement of charge into and out of the cell, thereby regulating neuron firing. One extremely useful technique for investigating the biophysical properties of these channels is called patch clamp recording. In this method, neuroscientists place a polished glass micropipette against a cell and apply suction to form a high resistance seal. This process isolates a small “patch” of membrane that contains one or more ion channels.
