Ovarian Cortex Cryopreservation

Ovarian cortex cryopreservation is a fertility-preservation technique that stores ovarian cortical tissue at very low temperatures to maintain viable follicles for possible future use. The cortex contains many primordial follicles, and the process uses cryoprotective agents plus controlled slow cooling or vitrification to limit ice-crystal formation and cellular injury; tissue is later warmed and assessed for follicle survival and function. Ovarian cortex cryopreservation can preserve reproductive potential for patients facing gonadotoxic cancer treatments, ovarian surgery, or conditions that threaten ovarian reserve. Research in biology uses this approach to study follicle activation, tissue viability, and restoration of endocrine and reproductive activity.

This protocol describes a novel three-dimensional (3-D) culture model using a tyramine-linked hyaluronan hydrogel to encapsulate and culture preantral follicles from the mouse ovary. We also detail two approaches to ovarian follicle cryopreservation by vitrification.

Bovine Ovarian Cortex Tissue Culture

In vitro culture of bovine ovarian cortex and the effect of nutritional Stair-step diet on ovarian microenvironment is presented. Ovarian cortex pieces were cultured for seven days and steroids, cytokines, and follicle stages were evaluated. The Stair-Step diet treatment had increased steroidogenesis resulting in follicle progression in culture.

Mouse Sperm Cryopreservation and Recovery using the I·Cryo Kit

Here we demonstrate the newly developed I•Cryo kit for mouse sperm cryopreservation. Two-cell stage embryo development with frozen-thawed sperm was improved consistently in 5 mouse strains with the use of this kit. Over a 1.5 year period, 49 genetically modified mouse lines were archived by sperm cryopreservation with the I•Cryo kit and later successfully recovered by IVF.

Isolation and Cryopreservation of Neonatal Rat Cardiomyocytes

The isolation of neonatal rat cardiomyocytes is a time consuming and unpredictable procedure. This study describes methods for cryopreservation and thawing of neonatal rat cardiomyocytes that allows for more efficient use of cells. The thawed NRCMs can be used for various experiments without the need for performing isolations each time.
