Urothelial Barrier

The urothelial barrier is a specialized epithelial lining that protects the urinary tract by separating underlying tissues from urine and limiting the passage of water, ions, and potentially harmful solutes. Its function depends on multilayered urothelium, tight junctions between cells, and uroplakins concentrated in the apical membranes of umbrella cells, which maintain low permeability while allowing the tissue to stretch as the bladder fills. Studying this barrier helps explain urinary tract defense, bladder physiology, and diseases that disrupt epithelial integrity, including infection and inflammation, while supporting research into urothelial models, drug transport, and tissue repair.

The aim of the protocol is to present an optimized procedure for the establishment of an in vitro blood-brain barrier (BBB) model based on primary porcine brain endothelial cells (pBECs). The model shows high reproducibility, high tightness, and is suitable for studies of transport and intracellular trafficking in drug discovery.

Physiological Barriers

Physiological barriers are semi-permeable cellular structures restricting drug diffusion into intracellular compartments and tissues. There are six types of physiological barriers: blood endothelial, cell membrane, blood-brain, blood-cerebrospinal fluid (CSF), blood-placenta, and blood-testis barriers. The blood endothelial barrier is the most porous of these. It allows all small ionized, un-ionized, and lipophilic molecules to pass through the endothelial lining into the interstitial space...

To investigate the blood-retinal barrier permeability and the inner limiting membrane integrity in animal models of retinal disease, we used several adeno-associated virus (AAV) variants as tools to label retinal neurons and glia. Virus mediated reporter gene expression is then used as an indicator of retinal barrier permeability.
