Paxillin Talin Adaptors

Paxillin and talin adaptors are cytoskeletal scaffold proteins that organize integrin-based cell adhesions, enabling cells to attach to and sense the extracellular matrix. Talin binds the cytoplasmic tails of integrins and links them to actin filaments, while paxillin recruits signaling and structural proteins at focal adhesions; together, their interactions regulate adhesion assembly and force transmission. Studying these adaptors helps explain how cells control spreading, migration, and mechanosensing during development, wound repair, and tissue organization. Their dysfunction is also relevant to cancer invasion and other disorders involving abnormal cell adhesion, making them important targets in cell biology and mechanobiology research.

Among different methods to evaluate gene expression, the high-throughput sequencing of RNA, or RNA-seq. is particularly attractive, as it can be performed and analyzed without relying on prior available genomic information. During RNA-seq, RNA isolated from samples of interest is used to generate a DNA library, which is then amplified and sequenced. Ultimately, RNA-seq can determine which genes are expressed, the levels of their expression, and the presence of any previously unknown transcripts.

This video demonstrates a technique to study NOD-like receptor family pyrin domain-containing 3 (NLRP3) inflammasome formation in primary macrophages via immunofluorescence. The macrophages are first primed with bacterial lipopolysaccharide (LPS) to upregulate NLRP3 protein expression. Exposing the macrophages to nigericin—a potassium ionophore—leads to the activation of NLRP3 and the formation of inflammasomes, which are visualized via immunofluorescence.
