Pbs Washing Step

The PBS washing step is a laboratory procedure that rinses biological samples or cultured cells with phosphate-buffered saline to remove unwanted substances while preserving sample conditions. PBS provides a near-physiological ionic environment and stable pH, helping limit osmotic stress as unbound reagents, residual media, proteins, or debris are displaced and removed. In biology, this step supports reliable cell culture, immunostaining, microscopy, and molecular assays by reducing background and preventing carryover between protocol stages. Careful washing, including adequate coverage and gentle handling, can improve signal quality, sample integrity, and experimental reproducibility.

Washing, Drying, and Ignition of Precipitates

After filtration, the precipitate is washed to remove coprecipitated impurities and any remaining mother liquor. Colloidal precipitates, such as silver chloride, are washed with an electrolyte (such as dilute nitric acid) to prevent the peptization of the precipitate. In the case of slightly soluble precipitates, the wash solution contains a common ion to reduce solubility. Lead sulfate, which is slightly soluble in water, is washed with dilute sulfuric acid. Similarly, wash solutions may be...

Retinal pigment epithelium (RPE) replacement strategies and gene-based therapy are considered for several retinal degenerative conditions. For clinical translation, large eye animal models are required to study surgical techniques applicable in patients. Here we present a rabbit model for subretinal surgery geared towards RPE transplantation, which is versatile and cost-efficient.
