Phase Lock Gel

Phase Lock Gel is a specialized laboratory gel used to simplify liquid-liquid phase separation during biological sample processing. In a tube, centrifugation drives aqueous and organic solutions into distinct layers while the gel forms a stable barrier at their interface, preventing mixing and reducing cross-contamination during pipetting. This makes Phase Lock Gel useful for isolating nucleic acids and other biomolecules from complex samples, improving transfer efficiency and helping produce cleaner preparations for downstream analyses such as PCR, sequencing, and molecular biology workflows.

Self-Locking Screw

A square-threaded screw jack is a mechanical device widely used for lifting heavy loads or applying considerable force. One of the key features that can make a screw jack more effective and reliable is its self-locking capability. A square-threaded screw jack carrying a load is considered self-locking if the screw retains its position even after the moment applied to it is removed. This occurs when the frictional force direction reverses, and the reaction force acts on the other side of the...

Gel Purification

2023

Gel purification is used to recover DNA fragments after electrophoretic separation. DNA recovery from an agarose gel includes three basic steps: binding, washing and eluting from a silica column. DNA is believed to bind to silica in the presence of high salt via a salt bridge. Following binding, DNA is washed of impurities and eluted under low salt conditions disrupting this interaction. This video goes through a step-by-step, generalized procedure for cutting out a band from the gel, gel...

Phase Diagrams

A phase diagram combines plots of pressure versus temperature for the liquid-gas, solid-liquid, and solid-gas phase-transition equilibria of a substance. These diagrams indicate the physical states that exist under specific conditions of pressure and temperature and also provide the pressure dependence of the phase-transition temperatures (melting points, sublimation points, boiling points). Regions or areas labeled solid, liquid, and gas represent single phases, while lines or curves represent...

DNA Gel Electrophoresis

2023

DNA gel electrophoresis is a technique used for the detection and separation of DNA molecules. An electric field is applied to a gel matrix comprised of agarose, and within the gel, charge particles will migrate and separate based on size. The negatively charged phosphates of the DNA backbone cause DNA fragments to move toward the anode - a positively charged electrode. The video explains the mechanism by which DNA fragments are resolved on an agarose gel, and it provides a step-by-step...
