Porcine Aortic Endothelial Cell Isolation

Porcine aortic endothelial cell isolation is a laboratory technique for obtaining endothelial cells from the inner lining of pig aortas, providing a primary model for vascular biology and endothelial function. The procedure generally involves sterile removal and opening of the vessel, exposure of the luminal surface, and enzymatic or mechanical release of adherent cells, followed by collection and culture under conditions that support endothelial attachment and growth. Resulting cultures enable analysis of endothelial phenotype, barrier behavior, inflammatory responses, and interactions with blood-contacting materials. Because porcine vessels share important structural and physiological features with human arteries, this preparation supports cardiovascular research, vascular injury studies, tissue engineering, and translational biomaterials development.

Our protocol provides an effective means of identifying mechanisms in endothelial cell physiopathology.

Primary cell culture is a useful technique for analyzing specific populations of cells, particularly from transgenic mouse embryos at specific developmental stages. Herein, embryonic ventricles are dissected and dissociated, and antibody-conjugated beads recognize and separate out the endothelial cells for further analysis.
