Quadrant Streak Method

The Quadrant Streak Method is a microbiological technique used to separate and obtain isolated colonies from a mixed or concentrated population of microorganisms on solid growth medium. A sterile inoculating loop progressively spreads cells across four marked sections of an agar plate, sterilizing the loop between streaks to dilute the inoculum and reduce cell density in each successive quadrant. After incubation under suitable conditions, individual colonies can be examined and transferred to establish pure cultures for identification, biochemical testing, microscopy, or genetic analysis. This method supports routine bacterial cultivation, contamination assessment, and research that requires genetically or phenotypically consistent samples.

We describe here a behavioral setup and data analysis method for assaying olfactory responses of up to 100 vinegar flies (Drosophila melanogaster). This system may be used with single or multiple olfactory stimuli, and adaptable for optogenetic activation or silencing of neuronal subsets.
