Reichert Membrane Dissection

Reichert membrane dissection is a microsurgical technique used to isolate Reichert’s membrane, a specialized extracellular matrix layer that separates embryonic and extraembryonic tissues during early mammalian development. Under a stereomicroscope, fine instruments separate the membrane from the surrounding conceptus while preserving its structure for analysis. The isolated membrane can be examined for its composition, organization, and interactions with trophoblast and endoderm, providing insight into basement-membrane biology, implantation, tissue separation, and embryonic patterning. This preparation also supports studies of extracellular matrix function and developmental abnormalities in experimental biology.

Here we present a protocol for Ca2+ imaging in neurons and glial cells, which enables the dissection of Ca2+ signals at subcellular resolution. This process is applicable to all cell types that allow the expression of genetically encoded Ca2+ indicators.

Use of a Robot for High-throughput Crystallization of Membrane Proteins in Lipidic Mesophases

Herein is described a robotic approach to high-throughput crystallization of membrane proteins in lipidic mesophases for use in structure determination using macromolecular X-ray crystallography. Three robots capable of handling the viscous and sticky protein-laden mesophase integral to the method are introduced.

Immunostaining of Dissected Zebrafish Embryonic Heart

A rapid way to conduct immunostaining of zebrafish embryonic heart is described. Compared to the whole mount immunostaining approach, this method dramatically increases the penetration of the antibodies, which allows obtaining high resolution images that reveal cellular/subcellular structures in the heart within a much reduced processing time.
