Rna Directionality

RNA directionality is the chemical polarity that gives every RNA strand distinct 5′ and 3′ ends, a feature that determines how genetic information is read and processed. This polarity arises from phosphodiester bonds linking the 5′ phosphate of one ribose to the 3′ hydroxyl of the next; RNA polymerases therefore synthesize strands from 5′ to 3′ while reading a complementary template in the opposite direction. Directionality guides transcription, translation, RNA splicing, and degradation by establishing how enzymes and ribosomes bind and move along RNA molecules. It is also essential for interpreting gene orientation, designing primers and probes, and analyzing RNA sequencing data.

Direct Restart of a Replication Fork Stalled by a Head-On RNA Polymerase

The fate of the replisome following a collision with a head-on RNA polymerase (RNAP) is unknown. We find that the replisome stalls upon collision with a head-on RNAP, but resumes elongation after displacing the RNAP from DNA. Mfd promotes replication restart by facilitating displacement of the RNAP after the collision.

This manuscript describes an approach to isolate select cognate RNPs formed in eukaryotic cells via a specific oligonucleotide-directed enrichment. We demonstrate the applicability of this approach by isolating a cognate RNP bound to the retroviral 5' untranslated region that is composed of DHX9/RNA helicase A.
