Sex Lethal Gene

The Sex-lethal gene is a key regulatory gene in Drosophila biology that determines sexual development and helps balance X-chromosome gene expression between XX females and XY males. It works through a dosage-sensitive early signal: in XX embryos, Sex-lethal transcripts are spliced to produce functional Sxl protein, which maintains its own female-specific splicing and regulates downstream targets, including alternative splicing of transformer and translation of msl-2. In XY embryos, this feedback is not established, leading to male-specific developmental and dosage-compensation pathways. The gene provides a powerful model for studying gene regulation, sex determination, and post-transcriptional control.

Shifting Zebrafish Lethal Skeletal Mutant Penetrance by Progeny Testing

The goal of this protocol is to alter the penetrance of lethal skeletal mutant phenotypes in zebrafish by selective breeding. Lethal mutants cannot be grown to adulthood and bred themselves, therefore this protocol describes a method for tracking and selecting penetrance through multiple generations by progeny testing.
