Stem-loop Unfolding

Stem-loop unfolding is the disruption of a nucleic acid hairpin, a secondary structure formed when complementary sequences within a single strand pair to create a stem and leave a loop. Unfolding occurs when heat, chemical conditions, mechanical force, or proteins such as helicases destabilize base pairing, allowing the paired regions to separate and the strand to adopt a more extended conformation. In biology, studying stem-loop unfolding reveals how RNA structure regulates translation, transcription, splicing, and molecular recognition. Experimental analysis also supports the design of biosensors, nucleic acid probes, and single-molecule assays that measure folding stability and structural transitions.

Neural Activity Propagation in an Unfolded Hippocampal Preparation with a Penetrating Micro-electrode Array

We have developed an in vitro unfolded hippocampus which preserves CA1-CA3 array of neurons. Combined with the penetrating micro-electrode array, neural activity can be monitored in both the longitudinal and transverse orientations. This method provides advantages over hippocampal slice preparations as the propagation in the entire hippocampus can be recorded simultaneously.
