Subcellular Mrna Localization

Subcellular mRNA localization is the directed positioning of messenger RNA molecules within distinct regions of a cell, a process that enables proteins to be produced where they are needed. Localization typically depends on sequence elements within the mRNA, RNA-binding proteins, transport along the cytoskeleton, and anchoring at specific cellular sites, where local translation can occur. This spatial regulation supports cell polarity, development, neuronal function, and tissue organization by coordinating gene expression with cellular architecture. Studying mRNA localization helps researchers understand how cells control protein distribution and how disruptions in RNA transport or local translation may contribute to disease.

An Immunolabeling Technique to Visualize the Subcellular Localization of Proteins

This video demonstrates a technique for the immunolabeling of sigma-1 receptors in a murine heart tissue section using quantum dots. The heart tissue is fixed, osmicated, stained, dehydrated, embedded in resin, and then sectioned. The sections are then treated with antibodies and labeled with quantum dots to demarcate the subcellular localization of Sigma-1 receptors using electron microscopy.

Labeling the extracellular domain of a membrane protein with a pH sensitive fluorophore, superecliptic pHluorin (SEP), allows subcellular localization, expression, and trafficking to be determined. Imaging SEP-labeled proteins with total internal reflection fluorescence microscopy (TIRFM) enables the quantification of protein levels in the peripheral ER and plasma membrane.
