Thp-1 Cells

THP-1 cells are a human monocytic leukemia cell line widely used as an in vitro model of monocytes and macrophages in immunology and cell biology. These suspension-growing cells can be differentiated into adherent, macrophage-like cells, commonly through treatment with phorbol 12-myristate 13-acetate (PMA), which activates protein kinase C signaling and promotes changes in morphology, adhesion, and immune function. Researchers use THP-1 cells to study innate immune responses, inflammation, cytokine production, host-pathogen interactions, and responses to candidate therapeutics. Their standardized growth and manipulability support reproducible experiments, although results should be interpreted alongside findings from primary human cells.

This protocol is based on LPS and ATP-induced death of PMA-differentiated THP-1 macrophages. We use flow cytometry to analyze Annexin V and 7-AAD double staining to detect cell death, using the whole cell and employing scanning electron microscopy to observe the cell membrane morphology.
