Tissue Culture Preparation

Tissue culture preparation is the set of steps used to obtain and condition biological tissue for growth and maintenance outside the organism. In biology, this process typically combines careful collection, trimming or dissociation of tissue, and transfer into sterile culture vessels containing a suitable nutrient medium. Aseptic technique limits contamination, while controlled temperature, gas exchange, and medium conditions support cell survival and preserve tissue structure or function. Prepared cultures provide a foundation for studying cell behavior, development, disease, toxicity, and regenerative processes, and can support primary cultures, explant models, and tissue engineering research.

Preparation of Primary Mixed Glial Cultures from Adult Mouse Spinal Cord Tissue

The development of neuropathic pain involves pathological changes of spinal cord glial cells. A reliable glial culture system derived from adult spinal cord tissue and designed for studying these cells in vitro is lacking. Therefore, we show here how to establish primary mixed glial cultures from adult mouse spinal cord tissue.

Histotypic Tissue Culture

Although two-dimensional tissue culture has been common for some time, cells behave more realistically in a three-dimensional culture, and more closely mimics native tissue. This video introduces histotypic tissue culture, where the growth and propagation of one cell line is done in an engineered three-dimensional matrix to reach high cell density. Here, we show the harvesting of cells from donor tissue, followed by cell culture on an engineered construct.

Whole Organ Tissue Culture

Whole organs can be cultured ex vivo using specialized bioreactors with the goal of repairing or replacing entire organs. This method uses a donor organ that is stripped of all cells, leaving behind the three-dimensional structure, which is then repopulated with new cells. This video demonstrates the whole organ culture of lungs and shows how a dynamic culture that mimics the mechanical stimulation in the body is needed to induce native tissue properties.

The procedure of preparing slices containing the adult mouse hypothalamic suprachiasmatic nucleus (SCN), and a rapid way to culture the SCN tissue in organotypic culture condition, are reported. Further, the measurement of oscillatory clock gene protein expression using dynamic luciferase reporter technology is described.
