Nkt Cell Analysis

NKT cell analysis is the characterization of natural killer T cells, a specialized lymphocyte population that combines T-cell receptor recognition with natural killer cell functions. Researchers identify these cells by measuring markers such as CD3 and CD56, assessing CD1d-dependent recognition of lipid antigens, and evaluating rapid cytokine release or cytotoxic activity. Flow cytometry, antigen-specific stimulation, and functional assays can reveal NKT cell frequency, phenotype, and responses in biological samples. These measurements support studies of immune regulation, infection, cancer, and autoimmune disease, while helping researchers assess how NKT cells contribute to inflammation, tumor surveillance, and potential immunotherapies.

Here we describe a method for activating and expanding human NKT cells from bulk T cell populations using artificial antigen presenting cells (aAPC). The use of CD1d-based aAPC provides a standardized method for generating high numbers of functional NKT cells.

Determining the cell cycle position of a population of cells, or understanding how signals affect proliferation, can be readily measured by flow cytometry using this protocol. We report a simple experimental approach to staining cells and quantifying their position in the cell cycle.

Transcriptome Analysis of Single Cells

In this article we describe a simple method for the harvesting of single cells from rat primary neuronal cultures and subsequent transcriptome analysis using aRNA amplification. This approach is generalizable to any cell type.
