Transient Plant Expression

Transient plant expression is a technique for producing a target protein or RNA in plant cells for a limited period without permanently integrating the introduced genetic material into the plant genome. It typically involves delivering a gene construct through Agrobacterium-mediated infiltration, particle bombardment, or another transformation method, after which host-cell machinery transcribes and translates the construct before its activity declines as the material is degraded or diluted. This rapid, flexible approach supports gene-function studies, protein production, vaccine and antibody research, and evaluation of engineered constructs. Compared with stable transformation, transient expression can shorten experiments and enable testing of multiple designs without generating heritable plants.

Plants offer a novel system for the production of pharmaceutical proteins on a commercial scale that is more scalable, cost-efficient and safe than current expression paradigms. In this study, we report a simple and convenient, yet scalable approach to introduce target-gene containing Agrobacterium tumefaciens into plants for protein transient expression.

Tobacco plants were used to produce a fungal cellulase, TrCel5A, via a transient expression system. The expression could be monitored using a fluorescent fusion protein, and the protein activity was characterized post-expression.
