Trap-seq

TRAP-seq (Translating Ribosome Affinity Purification sequencing) is a cell-type-specific method for profiling messenger RNAs associated with actively translating ribosomes, helping researchers measure gene expression in defined populations within complex tissues. The approach genetically directs target cells to produce an epitope-tagged ribosomal protein; tissue lysates are prepared, tagged ribosomes are captured by affinity purification, and their associated mRNAs are isolated and analyzed by RNA sequencing. By enriching transcripts from selected cells without requiring physical cell separation, TRAP-seq supports studies of tissue heterogeneity, neural circuits, development, and disease. Its results reveal cell-specific translational programs and molecular changes linked to biological function.

Among different methods to evaluate gene expression, the high-throughput sequencing of RNA, or RNA-seq. is particularly attractive, as it can be performed and analyzed without relying on prior available genomic information. During RNA-seq, RNA isolated from samples of interest is used to generate a DNA library, which is then amplified and sequenced. Ultimately, RNA-seq can determine which genes are expressed, the levels of their expression, and the presence of any previously unknown transcripts.
