Tunica Vaginalis

The tunica vaginalis is a serous membrane surrounding the testis and epididymis, helping protect these structures within the scrotum. Derived from the embryonic processus vaginalis, it forms visceral and parietal layers separated by a potential cavity containing a small amount of lubricating fluid; this arrangement allows the testis to move smoothly while reducing friction. In biology and anatomy, the tunica vaginalis provides a model for understanding serous membranes, embryonic development, and male reproductive organization. Abnormal fluid accumulation between its layers causes a hydrocele, while trauma or inflammation can produce blood or inflammatory fluid in the cavity, making the membrane clinically significant.

A mouse model of uropathogenic E. coli (UPEC) transurethral inoculation to establish latent intracellular bladder reservoirs and subsequent bladder exposure to G. vaginalis to induce recurrent UPEC UTI is demonstrated. Also demonstrated are the enumeration of bacteria, urine cytology, and in situ bladder fixation and processing for scanning electron microscopy.

Protocols for germ cell transplantation and testis tissue xenografting are described. Germ cell transplantation results in donor-derived spermatogenesis in recipient testes and represents a functional reconstitution assay for identification of spermatogonial stem cells (SSCs). Testis tissue xenografting reproduces testis development and spermatogenesis of various donor species in recipient mice.
