Viral Pellet Resuspension

Viral pellet resuspension is a laboratory technique used to recover concentrated virus particles collected as a pellet after centrifugation, enabling further analysis or processing. After the supernatant is removed, the pellet is gently mixed with an appropriate buffer so liquid penetrates the compacted material and releases virions into a uniform suspension while minimizing loss or damage. The resulting viral preparation can support infectivity measurements, molecular assays, purification workflows, and other biology experiments that require a defined virus concentration. Careful handling during resuspension improves sample consistency and helps preserve viral activity for downstream applications.

Preparation of Viral DNA from Nucleocapsids

We describe the process of isolating high purity herpesvirus nucleocapsid DNA from infected cells. The final DNA captured from solution is of high concentration and purity, making it ideally suited for high-throughput sequencing, high fidelity PCR reactions, and transfections to produce new viral recombinants.

In this study, a protocol was developed to produce good quality pellets using a flat die pellet mill at reduced specific energy consumption testing high-moisture corn stover and a starch based binder. The results indicated that adding a corn starch binder improved the pellet durability, reduced percent fines and decreased specific energy consumption.
