Zinc Transport Kinetics

Zinc transport kinetics describes how quickly and efficiently zinc moves across biological membranes, providing a quantitative view of zinc homeostasis in cells and organisms. Transport proteins mediate zinc influx, efflux, and intracellular sequestration, while concentration gradients, transporter abundance, binding affinity, and saturation influence rates that can be analyzed using kinetic models. In biology, these measurements clarify how cells maintain zinc levels needed for enzyme activity, gene regulation, and signaling without permitting toxic accumulation. Comparing transport kinetics across tissues, organisms, or experimental conditions supports research on nutrient regulation, metal toxicity, immune function, and diseases linked to disrupted zinc balance.

Zinc transport has proven challenging to measure due to the weak causal links to protein function and the low temporal resolution. This protocol describes a method for monitoring, with high temporal resolution, Zn2+ extrusion from living cells by utilizing a Zn2+ sensitive fluorescent dye, thus providing a direct measure of Zn2+ efflux.

(2018)This video demonstrates a whole-cell electrochemical method to assess how proton transfer affects extracellular electron transport (EET) in electroactive bacteria. By replacing protons with deuterons using deuterium oxide, the method measures changes in current to reveal a kinetic isotope effect, highlighting the role of...

Kinetic Energy

2019

Kinetic energy is the ability of an object in motion to do work or enact change. It can take on many forms. For instance, water flowing down a waterfall has kinetic energy. In biological systems, particles of light travel and are absorbed by plants to create chemical energy. Animals consume the chemical energy and give off molecules that carry their scent through the air. They also generate kinetic energy when they run away from predators. Entire systems also possess kinetic energy, like the...

Enzyme Kinetics

Enzymes speed up reactions by lowering the activation energy of the reactants. The speed at which the enzyme turns reactants into products is called the rate of reaction. Several factors impact the rate of reaction, including the number of available reactants. Enzyme kinetics is the study of how an enzyme changes the rate of a reaction. Scientists typically study enzyme kinetics with a fixed amount of enzyme in the controlled environment of a test tube. When more reactant, or substrate, is...
