Lamina Propria Mononuclear Cells

Lamina propria mononuclear cells are immune cells isolated from the connective tissue beneath mucosal epithelium, particularly in the gastrointestinal tract, where they regulate tissue defense and inflammation. Researchers typically remove the epithelium, enzymatically digest the underlying tissue, and separate the released leukocytes to obtain populations such as lymphocytes, macrophages, and dendritic cells. In cancer research, these cells provide a window into the tumor microenvironment and mucosal immune responses, enabling analysis of immune-cell infiltration, cytokine production, antigen recognition, and interactions with tumor cells. Their study supports investigation of colorectal cancer development, immunotherapy responses, and mechanisms of tumor-associated inflammation.

The goal of this protocol is to isolate mononuclear cells that reside in the lamina propria of the colon by enzymatic digestion of the tissue using collagenase. Here, we describe the isolation of enteric-glial cells from the intestinal-submucosa using sequential EDTA incubations to chelate divalent cations and then incubation in non-enzymatic cell recovery solution. Plating the resultant cell suspension on poly-D-lysine and laminin results in a highly enriched culture of submucosal glial cells for functional analysis.
