Mouse Tumor Samples

Mouse tumor samples are cancerous tissues or cells collected from mice bearing tumors and are important for examining tumor biology in a controlled research model. Researchers preserve these samples through procedures such as fixation, freezing, or processing into cell suspensions, allowing microscopic, molecular, and cellular analyses of tumor structure, gene activity, and interactions with surrounding tissue. In cancer research, mouse tumor samples help evaluate tumor growth, treatment responses, biomarkers, and drug effects, while comparisons across experimental groups can connect laboratory findings with disease mechanisms and guide the development of potential therapies.

Tumor Implantation and Injection of a Tumor Vaccine in a Mouse Model

This video demonstrates a cell-based tumor vaccination assay. A mouse, intradermally injected with B16-F10 melanoma cells for tumor formation, is vaccinated with B16-F10 cells secreting Flt3L cytokine near the developed tumor. The secreted Flt3L from the vaccine promotes the maturation and proliferation of dendritic cells that then present tumor antigens to T cells in lymph nodes, triggering a targeted anti-tumor immune response.

Begin with immunodeficient mice bearing subcutaneous xenograft tumors derived from human colorectal cancer cells.Orally administer Pediococcus pentosaceus, a lactic acid bacterium safe for consumption and engineered to secrete an anticancer protein, to the treated mouse.Administer saline to the control mouse.Being acid-tolerant, the bacteria survive the acidic environment of the stomach and reach the intestine. There, they colonize the intestinal lumen and secrete the anticancer protein.The...

Orthotopic Homograft Tumor Transplantation: A Technique to Generate Pancreatic Tumor Mouse Models

The video explains a stepwise surgical method for orthotopic implantation of a tumor fragment from a cancerous donor to the pancreas of a non-cancerous recipient mice for development of pancreatic cancer. Orthotopically implanted PDAC could recapture the tissue microenvironment of human pancreatic cancers than conventional subcutaneous (SC) models more accurately.

Source: Alomari, S., et al. Implementation of Minimally Invasive Brain Tumor Resection in Rodents for High Viability Tissue Collection. J. Vis. Exp. (2022)This video demonstrates the use of a minimally invasive resection system (MIRS) to excise tumor tissue from an anesthetized mouse implanted with a tumor. The procedure involves securing the mouse onto a stereotactic frame, reopening the previous surgical site to access the brain, and inserting a specialized cannula that utilizes suction and a...
