Orthotopic Allograft Implantation

Orthotopic allograft implantation is a preclinical transplantation technique in which tumor cells or tissue from a donor of the same species are placed into the corresponding organ of a recipient, providing a model that more closely reflects cancer in its native anatomical setting. The graft is surgically positioned within the organ, allowing interactions with local stromal cells, blood vessels, extracellular matrix, and, when applicable, the host immune system to influence tumor growth and dissemination. In cancer research, this approach supports the study of tumor progression, invasion, metastasis, and treatment response, helping investigators evaluate disease mechanisms and therapeutic strategies in physiologically relevant models.

Syngeneic mouse orthotopic allografts of pancreatic ductal adenocarcinoma (PDAC) recapitulate the biology, phenotypes, and therapeutic responses of disease subtypes. Owing to their fast, reproducible tumor progression, they are widely used in preclinical studies. Here, we show common practices to generate these models, injecting syngeneic murine PDAC cultures into the pancreas.

The rat orthotopic renal transplantation model contributes to investigating the mechanism of renal allograft rejection. The current model increases the recipients' survival without interference with blood supply and venous reflux of the lower body using an end-to-end anastomosis of kidney implantation and an end-to-side "tunnel" method of ureter-bladder anastomosis.
