Patient-matched Fibroblasts

Patient-matched fibroblasts are connective-tissue cells obtained from the same individual as a tumor or cancer model, providing a personalized representation of the patient’s stromal environment. Researchers isolate these cells from patient tissue, expand them under controlled culture conditions, and combine them with cancer cells or three-dimensional models to examine cell-to-cell signaling, extracellular-matrix remodeling, and changes in tumor behavior. In cancer research, patient-matched fibroblasts help reveal how the tumor microenvironment influences invasion, growth, treatment response, and resistance. They also support more representative drug-testing systems and may improve the development of personalized therapeutic strategies.

Carcinoma-associated fibroblasts (CAFs) rich in myofibroblasts present within the tumour stroma, play a major role in driving tumour progression. We developed a coimplantation tumour xengraft model for experimentally generating CAFs from human mammary fibroblasts. The protocol describes how to establish CAF myofibroblasts that acquire an ability to promote tumourigenesis.
