Peripheral Blood Nk

Peripheral blood NK cells are natural killer lymphocytes circulating in the bloodstream that provide rapid immune surveillance against abnormal cells, including cancer cells. They recognize stress-associated or altered surface signals without prior antigen sensitization and respond by releasing perforin and granzymes, which trigger target-cell death, while also producing cytokines that shape immune activity. In cancer research, these cells serve as accessible models for studying innate antitumor immunity, tumor-cell recognition, and immune evasion. Their isolation, functional testing, and expansion support biomarker development and the evaluation of NK cell-based immunotherapies, including approaches designed to improve persistence and tumor targeting.

Natural killer cells are a small population of lymphocytes. Here we show how to isolate these cells from human blood by negative selection, using a kit from StemCell Technologies. The cells obtained are viable and untouched by antibodies, and therefore ready to be used for a number of procedures.

Ex Vivo Expansion of Natural Killer Cells from Peripheral Blood Mononuclear Cells

This video demonstrates a technique for the expansion of natural killer (NK) cells directly from peripheral blood mononuclear cells (PBMCs) using a feeder cell line. PBMCs are co-cultured with genetically modified lymphoblastoid feeder cells expressing membrane-bound interleukin-21 (mIL-21), and the culture is supplemented with the cytokines interleukin-2 (IL-2) and interleukin-15 (IL-15). Together, this cocktail of ligands leads to the prolonged expansion of NK cells with high purity.

Dextran Enhances the Lentiviral Transduction Efficiency of Murine and Human Primary NK Cells

The goal of this study was to formulate technologies that allow for successful gene transduction in primary natural killer (NK) cells. The dextran-mediated lentiviral transduction of human or mouse primary NK cells results in higher gene expression efficiencies. This method of gene transduction will vastly improve NK cell genetic manipulation.

Expansion of Human Peripheral Blood γδ T Cells using Zoledronate

A method to expand γδ T cells from peripheral blood mononuclear cells (PBMC) is described. PBMC-derived γδ T cells are stimulated and expanded using zoledronate and interleukin-2 (IL-2). Large scale expansion of γδ T cells can be applied to autologous cellular immunotherapy of cancer.
