Pre-metastatic Niche

The pre-metastatic niche is a tumor-conditioned microenvironment that forms in distant organs before cancer cells arrive, creating conditions that support metastatic colonization. Primary tumors release soluble factors, extracellular vesicles, and cytokines that travel through the circulation, recruit bone marrow-derived cells, remodel the extracellular matrix, and alter local immune activity in target tissues. Studying this process helps explain why certain organs become preferred sites for metastasis and reveals early stages of cancer dissemination. Pre-metastatic niche research supports the development of biomarkers for metastatic risk and therapies designed to prevent or disrupt the tissue changes that enable secondary tumor growth.
