Prognostic Signature

A prognostic signature is a pattern of molecular, clinical, or imaging features used to estimate a patient’s likely disease course, such as survival, recurrence, or treatment response. In cancer research, investigators combine measurements from tumors or patients and use statistical or computational models to identify features associated with outcomes, then validate the resulting signature in independent groups. Prognostic signatures can stratify patients into risk categories, support study design, and clarify biological differences among cancers. When rigorously validated, they may improve patient counseling and help guide surveillance or treatment decisions, while also providing a framework for developing more precise cancer research and care.

Here, a protocol is presented for optically extracting and cataloging innate cellular fluorescence signatures (i.e., cellular autofluorescence) from every individual live cell distributed in a three-dimensional space. This method is suitable for studying the innate fluorescence signature of diverse biological systems at a single-cell resolution, including cells from bacteria, fungi, yeasts, plants, and animals.

A structured protocol is presented for a cell-based assay as a functional test to predict the prognosis of idiopathic scoliosis using cellular dielectric spectroscopy (CDS). The assay can be performed with fresh or frozen peripheral blood mononuclear cells (PBMCs) and the procedure is completed within 4 days.
