Soft Agar Assay

The soft agar assay is an in vitro method for measuring anchorage-independent cell growth, a property often associated with malignant transformation and tumorigenic potential. Cells are suspended in a semisolid agar layer over a firmer agar base, preventing attachment to a rigid surface; cells capable of proliferating without adhesion form visible colonies during incubation. In cancer research, colony formation provides a functional readout of oncogene activity, transformation, and differences in tumor cell behavior. Researchers use the assay to compare cell lines, evaluate genetic or pharmacological interventions, and assess how treatments affect cancer cell proliferation under conditions that model loss of normal growth controls.

The soft agar colony formation assay is a method used to confirm cellular anchorage-independent growth in vitro. The goal of this protocol is to illustrate a stringent method for the detection of the tumorigenic potential of transformed cells and the tumor suppressive effects of proteins on transformed cells.

(2017)This video demonstrates the procedure for detecting bacteriocin-mediated killing by spotting supernatant from an antibiotic-induced donor strain onto a soft agar overlay containing a test strain. Clear zones formed after incubation indicate bacterial inhibition.
