Spheroid Growth Conditions

Spheroid growth conditions are the defined physical, chemical, and biological parameters used to form and maintain three-dimensional cell aggregates in culture, providing a model that more closely reflects tissue organization than many two-dimensional systems. In cancer research, cells are typically encouraged to assemble under nonadherent or low-attachment conditions, where limited substrate attachment promotes cell-cell interactions and compaction into spheroids. Control of cell density, culture medium, extracellular matrix support, oxygen availability, and incubation time influences spheroid size, architecture, viability, and internal gradients. These models help researchers study tumor growth, drug penetration, treatment response, invasion, and microenvironmental effects, supporting more physiologically relevant preclinical testing.

The uneven loss of medium from microtiter plates affects the reproducibility of uniform multicellular tumor spheroid formation. Improving culture conditions to reduce significant medium loss will improve the reproducibility of spheroid formation and the results of spheroid-based assays using the liquid-overlay technique.

The following protocol provides techniques for encapsulating pancreatic β-cells in step-growth PEG-peptide hydrogels formed by thiol-ene photo-click reactions. This material platform not only offers a cytocompatible microenvironment for cell encapsulation, but also permits user-controlled rapid recovery of cell structures formed within the hydrogels.
