Thermosensitive Liposomal Doxorubicin

Thermosensitive liposomal doxorubicin is a temperature-responsive drug-delivery formulation that encapsulates the anticancer agent doxorubicin within lipid vesicles, helping control where and when the drug is released. When exposed to localized mild hyperthermia, the liposomal membrane undergoes a phase transition that increases its permeability and rapidly releases doxorubicin near heated tumor tissue. In cancer research, this approach combines chemotherapy with thermal treatment to improve drug deposition at the target site while limiting exposure to healthy tissues. Researchers use it to study tumor targeting, heat-controlled release, treatment response, and strategies for improving the therapeutic index of doxorubicin.

Reconstituting functional membrane proteins into giant liposomes of defined composition is a powerful approach when combined with patch-clamp electrophysiology. However, conventional giant liposome production may be incompatible with protein stability. We describe protocols for producing giant liposomes from pure lipids or small liposomes containing ion channels.

The goal of this protocol is to use cationic/anionic liposomes with a neuro-targeting peptide as a CNS delivery system to enable siRNA to cross the BBB. The optimization of a delivery system for treatments, like siRNA, would allow for more treatment options for prion and other neurodegenerative diseases.

Monitoring B Cell Activation with Antigenic Liposomes using a Calcium-Flux Assay

2025

This video showcases the assessment of B cell activation through the utilization of antigenic liposomes. B cells treated with calcium-sensitive Indo-1 dye are distinguished using flow cytometry, and their violet-to-blue fluorescence ratio is analyzed. As the cells encounter antigenic liposomes, it triggers B cell activation through calcium influx, progressively enhancing the violet-to-blue fluorescence signal ratio over time.
