Tumor Cell Culture

Tumor cell culture is the controlled growth and maintenance of cancer cells outside the body, providing a laboratory model for studying tumor biology. Researchers sustain cells in nutrient-rich media under defined temperature and gas conditions, using adherent or suspension systems and periodic passaging to support proliferation while limiting contamination and unwanted changes. These cultures help investigators examine cell growth, signaling, invasion, and responses to anticancer treatments. By enabling controlled experiments and comparison of tumor cell behaviors, tumor cell culture supports cancer research, drug screening, biomarker studies, and the development of more effective therapeutic strategies.

This video demonstrates a method to establish a three-dimensional tumor spheroid and T cell co-culture. An agarose cast with microwells containing tumor spheroids is prepared, followed by the addition of T cells. After incubation, this setup allows studying the interaction of T cells with tumor spheroids in a controlled three-dimensional environment.

Organotypic hippocampal slice cultures (OHSC) represent an in vitro model that simulates the in vivo situation very well. Here we describe a vibratome-based improved slicing protocol to obtain high quality slices for use in assessing the neuroprotective potential of novel substances or the biological behavior of tumor cells.

Generating an Organoid Culture from a Brain Tumor Cell Suspension

This video describes the preparation and cultivation of brain tumor organoids from single-cell suspension. Using a laminin-rich extracellular matrix as a scaffold and culture media for nutrients, cells are facilitated to multiply and self-organize within the organoid. Consequently, ex vivo organoids are generated, mimicking cellular diversity and the microenvironment of brain tumors.
