Glow Stick Mechanism

The glow stick mechanism is a chemiluminescent process in which a chemical reaction produces visible light without generating substantial heat. Bending a glow stick breaks an inner glass vial, allowing hydrogen peroxide to mix with an oxalate ester; the reaction forms a high-energy intermediate that transfers energy to a fluorescent dye, which releases that energy as colored light. The glow intensity and duration depend on reaction conditions, especially temperature and reagent concentrations. Studying glow sticks provides an accessible model for reaction kinetics, energy transfer, fluorescence, and the design of portable, battery-free light sources.

High-throughput Functional Screening using a Homemade Dual-glow Luciferase Assay

We present a rapid and inexpensive screening method for identifying transcriptional regulators using high-throughput robotic transfections and a homemade dual-glow luciferase assay. This protocol rapidly generates direct side-by-side functional data for thousands of genes and is easily modifiable to target any gene of interest.

Finger-stick Blood Sampling Methodology for the Determination of Exercise-induced Lymphocyte Apoptosis

Exercise is capable of inducing apoptosis in immune cells. There are various measurement limitations, particularly relating to the amount of time required to isolate and treat a blood sample prior to the assessment. Demonstrated is a rapid and minimally invasive procedure for the analysis of exercise-induced lymphocyte apoptosis.

The biosynthesis of cartilaginous extracellular matrix by chondrocytes can be affected by application of mechanical stimuli. This method describes the technique of applying dynamic compressive strains to chondrocytes encapsulated in 3D constructs and the evaluation of induced changes in chondrocyte metabolism.

In this protocol, we demonstrate the fabrication of a microactuator array of vertically displaced posts on which the technology is based, and how this base technology can be modified to conduct high-throughput mechanically dynamic cell culture in both two-dimensional and three-dimensional culture paradigms.
