Hplc-free Synthesis

HPLC-free synthesis refers to chemical synthesis workflows that produce and isolate target compounds without using high-performance liquid chromatography for purification. Instead, these methods rely on properties such as solubility, polarity, phase partitioning, and selective reactivity to separate products through extraction, precipitation, crystallization, filtration, or other scalable operations. In chemistry, HPLC-free synthesis can simplify process development, reduce solvent and equipment requirements, and improve compatibility with larger-scale production. The approach is particularly useful when developing practical synthetic routes, where efficient isolation, reproducibility, and reduced processing time are as important as chemical yield and product purity.

Here, we describe in great detail an established and robust protocol for the extraction of glucosinolates from ground plant materials. After an on-column sulfatase treatment of the extracts, the desulfoglucosinolates are eluted and analyzed by high-pressure liquid chromatography.
