One-pot Synthesis

One-pot synthesis is a chemical strategy in which multiple reaction steps occur in the same vessel without isolating or purifying intermediate products, improving efficiency and reducing material use. The process typically uses sequential reagent addition or controlled changes in temperature, solvent, pH, or atmosphere so that each transformation proceeds under conditions compatible with the next. In chemistry, one-pot methods support the preparation of pharmaceuticals, natural products, polymers, and other complex molecules from simpler starting materials. By shortening workflows, limiting solvent and energy consumption, and reducing intermediate handling, they can improve overall yield, safety, and sustainability.

Microwave-assisted One-pot Synthesis of N-succinimidyl-4-[18F]fluorobenzoate ([18F]SFB)

A facile, one-pot synthesis of N-succinimidyl-4-[18F]fluorobenzoate ([18F]SFB) was developed based on a non-aqueous, three-step radiochemical process. Using microwave heating, the entire procedure can be completed in less than 30 min, or 60 min with further purification by preparative HPLC. The decay-corrected radiochemical yields (RCYs) were 35-5% (n > 30).
