Diffusion Capacity

Diffusion capacity is a pulmonary function measure that quantifies how efficiently gases move from the air in the alveoli into the bloodstream, providing important information about gas exchange. Clinically, it is commonly assessed with the diffusing capacity for carbon monoxide (DLCO), in which a small inhaled carbon monoxide concentration is measured after a breath-hold because carbon monoxide binds rapidly to hemoglobin; the result reflects the alveolar-capillary membrane and pulmonary capillary blood volume. Diffusion capacity testing helps evaluate interstitial lung disease, emphysema, pulmonary vascular disorders, and unexplained breathlessness, while also supporting disease monitoring, treatment assessment, and preoperative evaluation.

We describe a means to quickly and simply measure the lung diffusing capacity in mice and show that it is sufficiently sensitive to phenotype changes in multiple common lung pathologies. This metric thus brings direct translational relevance to the mouse models, since diffusing capacity is also easily measured in humans.

Cell Membranes and Diffusion In order to function, cells are required to move materials in and out of their cytoplasm via their cell membranes. These membranes are semipermeable, meaning that certain molecules are allowed to pass through, but not others. This movement of molecules is mediated by the phospholipid bilayer and its embedded proteins, some of which act as transport channels for molecules that otherwise would not be able to pass through the membrane, such as ions and carbohydrates.

Diffusion in Agar ExpandNOTE: In this exercise, you will be given agar containing an indicator chemical called phenolphthalein. When phenolphthalein is exposed to the normal alkaline conditions in the agar, it will look pink. But when it is exposed to neutral or acidic conditions, it changes from pink to clear. You will make different size and shaped agar cubes as a model for cells to study the impact of cell size and shape on diffusion rate. To make the first set of cells, measure out and cut...

Lung Capacity

The air in the lungs is measured in volumes and capacities. Lung volume measures reflect the amount of air taken in, released, or left over after a lung function, like a single inhalation. Lung capacity measures are sums of two or more lung volume measures. There are four defined lung volume measures: tidal volume, inspiratory reserve volume, expiratory reserve volume, and residual volume. Tidal volume is the amount of air inhaled and exhaled in a normal breath. Inspiratory...

This protocol presents a method to assess pulmonary alveolar-capillary reserve measured by combined single-breath measurement of the diffusing capacity to carbon monoxide (DL,CO) and nitric oxide (DL,NO) during exercise. Assumptions and recommendations for using the technique during exercise form the foundation of this article.
