Injection Drug Use

Injection drug use is the administration of psychoactive or other substances through a needle into a vein, muscle, or tissue, producing rapid systemic effects and important clinical consequences. By bypassing the gastrointestinal tract and delivering a substance directly into the bloodstream or surrounding tissue, injection can increase bioavailability, speed of action, and risk of overdose. Repeated injection may cause vascular injury, skin and soft-tissue infections, and transmission of blood-borne infections such as HIV and hepatitis C when equipment is shared. Clinical care emphasizes nonjudgmental screening, overdose prevention, sterile equipment access, infectious-disease testing, wound management, and evidence-based treatment for substance use disorders.

Supramolecular hydrogelators based on ureido-pyrimidinones allow full control over the macroscopic gel properties and the sol–gel switching behavior using pH. Here, we present a protocol for formulating and injecting such a supramolecular hydrogelator via a catheter delivery system for local delivery directly in relevant areas in the pig heart.

This protocol describes the fabrication of elastic 3D macroporous microcryogels by integrating microfabrication with cryogelation technology. Upon loading with cells, 3D microtissues are generated, which can be readily injected in vivo to facilitate regenerative therapy or assembled into arrays for in vitro high-throughput drug screening.
