Germline Dissection

Germline dissection is the isolation and examination of germline cells or reproductive tissues to understand how they develop and transmit genetic information across generations. In developmental biology, researchers separate germline components from surrounding somatic tissue and analyze their morphology, gene expression, cellular interactions, or developmental stage using microscopy and molecular or genetic methods. This approach can reveal how germline stem cells are maintained, how gametes form, and how signals between germline and soma shape reproductive development. By linking tissue organization with cell fate and inheritance, germline dissection supports studies of fertility, embryogenesis, developmental defects, and evolution.

We present an immunofluorescence imaging-based method for spatial and temporal localization of active ERK in the dissected C. elegans gonad. The protocol described here can be adapted for visualization of any signaling or structural protein in the C. elegans gonad, provided a suitable antibody reagent is available.
