Mouse Embryo Analysis

Mouse embryo analysis is the study of mouse embryos to characterize how cells, tissues, and organs form during development, making it a central approach in developmental biology. Researchers examine embryos at defined developmental stages using microscopy, tissue processing, genetic labeling, and molecular assays to track cell division, migration, differentiation, and tissue patterning. Comparing normal embryos with those carrying targeted genetic or environmental perturbations reveals how developmental pathways regulate body-plan formation and identifies causes of abnormal development. Because many mammalian processes are conserved, these analyses support research on gene function, congenital disorders, stem cell biology, and regenerative medicine.

This protocol presents a method to perform quantitative, single-cell in situ analysis of protein expression to study lineage specification in mouse preimplantation embryos. The procedures necessary for collection of blastocysts, whole-mount immunofluorescent detection of proteins, imaging of samples on a confocal microscope, and nuclear segmentation and image analysis are described.

Isolation of postimplantation-stage embryos allows one to study gene patterning and analyze cell-lineage decision making processes during embryonic development, but proper dissection of the early embryo can be challenging. This protocol describes a method for isolating early primitive-streak-stage embryos (~6.5 days post coitum [dpc]).
