Ploidy Analysis

Ploidy analysis is the determination of how many complete sets of chromosomes a cell or organism contains, a measurement that helps characterize genome organization and developmental state. It commonly estimates DNA content by staining cellular DNA and measuring fluorescence with flow cytometry, while chromosome-based methods can identify numerical changes directly in dividing cells. In developmental biology, comparing haploid, diploid, and polyploid cells or embryos reveals how genome duplication and chromosome imbalance affect cell proliferation, differentiation, fertility, and tissue formation. These measurements also support the study of embryogenesis, developmental abnormalities, and experimental models in which chromosome number is manipulated.

Manipulation of Ploidy in Caenorhabditis elegans

This method allows for the generation of tetraploid and triploid Caenorhabditis nematodes from any diploid strain. Polyploid strains generated by this method have been used to study chromosome interactions in meiotic prophase, and this method is useful for examining important basic questions in cell, developmental, evolutionary, and cancer biology.

Oocytes are prone to aneuploidy due to errors in chromosome segregation during meiotic maturation. Aneuploid eggs can cause infertility, miscarriages or developmental disorders like Down syndrome. Here, we describe methods to introduce materials of choice into oocytes and methods to study meiotic maturation and assess ploidy.

Chromosome Screening of Human Preimplantation Embryos by Using Spent Culture Medium: Sample Collection and Chromosomal Ploidy Analysis

The present study reports a protocol for chromosome screening of human embryos that uses spent culture medium, which avoids embryo biopsy and enables chromosome ploidy identification using NGS. The present article presents the detailed procedure, including the preparation of culture medium, whole genome amplification (WGA), next-generation sequencing (NGS) library preparation, and data analysis.

A High-Throughput In Situ Method for Estimation of Hepatocyte Nuclear Ploidy in Mice

We present a robust, cost-effective, and flexible method for measuring changes in hepatocyte number and nuclear ploidy within fixed/cryopreserved tissue samples that does not require flow cytometry. Our approach provides a powerful sample-wide signature of liver cytology ideal for tracking the progression of liver injury and disease.
