Borates

Borates are minerals and chemical compounds containing boron-oxygen groups, valued for their distinctive thermal, chemical, and structural properties in engineering materials. When heated, borate components can act as fluxes by lowering melting temperatures and promoting glass formation, while their ability to modify oxide networks influences viscosity, durability, and resistance to heat or chemical attack. These properties support applications in glass and ceramic manufacturing, enamels, thermal insulation, and cement-based materials. Understanding borate composition and phase behavior helps engineers tailor processing conditions and develop materials with improved strength, stability, and performance in demanding environments.

Here we present a protocol to express, solubilize, and purify several eukaryotic borate transporters with homology to the SLC4 transporter family using yeast. We also describe a chemical cross-linking assay to assess the purified homomeric proteins for multimeric assembly. These protocols can be adapted for other challenging membrane proteins.

Source: Al Ahmar, R., et al. Culture of Small Colony Variant of Pseudomonas aeruginosa and Quantitation of its Alginate. J. Vis. Exp. (2020)This video demonstrates a colorimetric assay to quantify alginate produced by Pseudomonas aeruginosa, using carbazole to detect uronic acids after acid hydrolysis. The method enables comparative analysis of alginate production normalized to bacterial biomass in research and clinical studies.

The bacterial cell wall is composed of peptidoglycan, a macromolecular network of sugar strands crosslinked by peptides. Ultra Performance Liquid Chromatography provides high resolution and throughput for novel discoveries of peptidoglycan composition. We present a procedure for the isolation of cell walls (sacculi) and their subsequent preparation for analysis via UPLC.
