Choked Flow Condition

Choked flow condition is a state in compressible-fluid systems where the mass flow rate reaches its maximum for a given upstream pressure, temperature, and flow passage. It occurs when the fluid accelerates to sonic velocity at the narrowest section, or throat, and the downstream-to-upstream pressure ratio falls below a critical value; further reducing downstream pressure cannot increase the flow rate because pressure disturbances cannot propagate upstream through the sonic region. Engineers use choked-flow analysis to size nozzles, orifices, and control valves, estimate mass-flow rates, and evaluate pressure-relief and propulsion systems. Understanding this condition supports safe, efficient design of gas-handling equipment and other high-speed flow systems.

This article first describes a procedure for isolating human endothelial cells from umbilical veins and then shows how to use these cells to examine neutrophil transmigration under flow conditions. By using a low-volume flow chamber made from a polymer with the optical characteristics of glass, live-cell fluorescent imaging of rare cell populations is also possible.

The protocol describes a reproducible method designed for use with cell culture supernatants to detect surface epitopes on small extracellular vesicles (EV). It utilizes specific EV immunoprecipitation using beads coupled with antibodies that recognize surface antigen CD9, CD63, and CD81. The method is optimized for downstream flow cytometry analysis.
