Decellularized Scaffold

Decellularized scaffolds are biomaterial frameworks made by removing cells and cellular components from tissues or organs while preserving much of the native extracellular matrix, making them important tools in tissue engineering. Chemical, enzymatic, or physical treatments disrupt cell membranes and wash away genetic material, while careful processing aims to retain matrix architecture, structural proteins, and biochemical cues. The resulting scaffold provides a three-dimensional environment for seeded cells, supporting adhesion, migration, proliferation, and tissue-specific organization. Engineers use decellularized scaffolds to study cell–matrix interactions and develop potential constructs for regenerative medicine, although source tissue, processing conditions, sterility, and immune compatibility strongly influence performance.

Perfusion decellularization is a novel technique to produce whole liver scaffolds that retains the organ's extracellular matrix composition and microarchitecture. Herein, the method of preparing whole organ scaffolds using perfusion decellularization and subsequent repopulation with hepatocytes is described. Functional and transplantable liver grafts can be generated using this technique.
