Sexing Mosquito Pupae

Sexing mosquito pupae is the identification and separation of male and female mosquitoes during the pupal stage, a practical step for genetics, colony management, and vector research. The method uses species-dependent sexual dimorphism, including differences in pupal size and the shape or position of terminal abdominal structures, which can be examined under a stereomicroscope before adult emergence. Accurate separation supports controlled crosses, sex-linked inheritance studies, and maintenance of defined laboratory strains, while also enabling experiments that require large numbers of males or females. Reliable pupal sexing can improve genetic screening and the production of male-only cohorts for vector-control research.

Aedes aegypti mosquitoes infected with Wolbachia are being released into natural populations to suppress the transmission of arboviruses. We describe methods to rear Ae. aegypti with Wolbachia infections in the laboratory for experiments and field release, taking precautions to minimize laboratory adaptation and selection.

RNA interference (RNAi) is an extremely valuable tool for uncovering gene function. However, the ability to target genes using RNAi during pre-adult stages is limited in the major human malaria vector Anopheles gambiae. We describe an RNAi protocol to reduce gene function via direct injection during pupal development.
