A549 Cell Infection

A549 cell infection is an in vitro method that uses human lung epithelial cells to model how pathogens interact with respiratory tissue, making it valuable for studying infection and host immunity. In a typical experiment, cultured A549 monolayers are exposed to a pathogen at a controlled multiplicity of infection, allowing adhesion, cellular entry, and, when supported, intracellular replication. Researchers then measure pathogen burden, cytopathic effects, and host responses such as inflammatory signaling or gene expression. This model supports investigations of respiratory pathogenesis, innate immune mechanisms, antiviral activity, and therapeutic interventions while providing a controlled complement to animal and clinical studies.

The video demonstrates a method to assess caspase-mediated protein cleavage in virus-infected cells. Post-viral infection, caspases get activated and degrade various host cellular proteins and viral nucleoproteins while specific caspase inhibitors prevent this degradation, as confirmed by western blotting.
