Automated Liquid Handler

An automated liquid handler is a laboratory instrument that uses programmable robotics to aspirate, dispense, and transfer liquids with consistent volume and timing, reducing repetitive manual pipetting. It works through software-controlled movement of probes or pipette tips, which draw samples and reagents from source vessels and deliver them to designated wells or tubes according to a defined protocol. In immunology and infection research, these systems support serial dilutions, assay setup, nucleic acid preparation, and high-throughput testing of patient or experimental samples. Standardized liquid handling can improve reproducibility, limit cross-contamination when appropriate safeguards are used, and increase throughput for workflows such as ELISA, PCR, and antimicrobial screening.

Here, an automated workflow to perform modular DNA "device" assembly using a modular cloning DNA assembly method on liquid-handling robots is presented. The protocol uses an intuitive software tool for generating liquid handler picklists for combinatorial DNA device library generation, which we demonstrate using two liquid handling platforms.

Whole blood Immunophenotyping is indispensable for monitoring the human immune system. However, the number of reagents required and the instability of specific fluorochromes in premixed cocktails necessitates daily reagent preparation. Here we show that semi-automated preparation of staining antibody cocktail helps establish reliable immunophenotyping by minimizing variability in reagent dispensing.
