Bl21(de3) Cells

BL21(DE3) cells are genetically engineered Escherichia coli commonly used to produce recombinant proteins, including antigens, antibodies, and other molecules relevant to immunology and infection research. Their chromosome carries the T7 RNA polymerase gene under control of the lacUV5 promoter, so adding an inducer such as IPTG triggers T7 polymerase production and strong transcription from plasmids containing a T7 promoter. Researchers introduce expression plasmids, optimize induction conditions, and harvest the resulting protein for purification and analysis. This system supports studies of pathogen proteins, host-pathogen interactions, immune recognition, and vaccine candidates, although protein folding, solubility, and endotoxin contamination require careful evaluation.

β-barrel outer membrane proteins (OMPs) serve many functions within the outer membranes of Gram-negative bacteria, mitochondria, and chloroplasts. Here, we hope to alleviate a known bottleneck in structural studies by presenting protocols for the production of β-barrel OMPs in sufficient quantities for structure determination by X-ray crystallography or NMR spectroscopy.

This protocol details the derivation of transplantable hematopoietic stem cells from mouse embryonic stem cells (ESC) and their subsequent injection into lethally irradiated recipient mice. Briefly, ESC are differentiated as embryoid bodies, which are then infected with retroviral HoxB4 and co-cultured with OP9 stromal cells and hematopoietic cytokines.
