Cell Culture Fluid

Cell culture fluid is the liquid environment that supports cells outside the body, providing the nutrients and conditions needed for their survival, growth, and specialized functions. Typically formulated with salts, sugars, amino acids, vitamins, buffering agents, and sometimes serum or defined growth factors, it maintains suitable pH, osmolarity, and energy availability while allowing cells to exchange metabolites and signaling molecules. In immunology and infection research, carefully selected culture fluids help maintain immune cells, expose them to pathogens or microbial products, and measure outcomes such as cytokine release, proliferation, cytotoxicity, and changes in host-pathogen interactions. Their composition strongly influences experimental reproducibility and interpretation.

Interstitial fluid flow is elevated in solid tumors and can modulate tumor cell invasion. Here we describe a technique to apply interstitial fluid flow to cells embedded in a matrix and then measure its effects on cell invasion. This technique can be easily adapted to study other systems.

A 3D System for Culturing Human Articular Chondrocytes in Synovial Fluid

A 3D system of culturing human articular chondrocytes in high levels of synovial fluid is described. Synovial fluid reflects the most natural microenvironment for articular cartilage, and can be easily obtained and stored. This system thus can be used for studying cartilage regeneration and for screening therapeutics for treating arthritis.

This protocol describes the reprogramming of primary amniotic fluid and membrane mesenchymal stem cells into induced pluripotent stem cells using a non-integrating episomal approach in fully chemically defined conditions. Procedures of extraction, culture, reprogramming, and characterization of the resulting induced pluripotent stem cells by stringent methods are detailed.
