Cell Mediated Lymphocytotoxicity

Cell-mediated lymphocytotoxicity is the immune process in which lymphocytes directly destroy infected, abnormal, or foreign target cells without requiring antibody-mediated killing. Cytotoxic T lymphocytes recognize antigen-derived peptides presented by major histocompatibility complex class I molecules, while natural killer cells respond to cellular stress or reduced self-signaling; both can trigger apoptosis through perforin and granzymes or death-receptor pathways. This activity helps control viral infections, eliminate emerging tumor cells, and contribute to transplant rejection. Laboratory cytotoxicity assays measure lymphocyte killing and support research into immune function, vaccine responses, cancer immunotherapy, and infection-related disease.

Lentiviral-mediated Knockdown During Ex Vivo Erythropoiesis of Human Hematopoietic Stem Cells

An ex vivo protocol to generate mature human red blood cells from hematopoietic stem/progenitors is described. Additionally we describe an efficient lentiviral-delivery method to knockdown the transcription factor TAL1 in primary erythroid cells. The efficiency of lentivirus mediated gene delivery is demonstrated using GFP expressing viruses.

Cell-mediated lymphocytotoxicity (CML) assays can be used to test autoreactive responses and study mechanisms of cell death in vitro. However, using live-cell confocal microscopic imaging techniques with fluorescent dyes, the type and kinetics of cell death as well as the pathways utilized can be studied in greater detail.

This video demonstrates the quantification of inflammatory mediators during infection by incubating human lung-tissue-derived immune cells with Haemophilus influenzae. Flow cytometry is used to measure cytokine fluorescence in labeled lymphocytes, correlating with the production of pro-inflammatory cytokines in response to bacterial infection.
