Competitive Repopulation Studies

Competitive repopulation studies are experimental assays that compare the ability of two or more cell populations to reconstitute or maintain an immune compartment under the same biological conditions. Researchers typically mix distinguishable donor cells, transfer them into a common recipient, and use genetic, phenotypic, or fluorescent markers to quantify each population over time; unequal representation reveals differences in survival, proliferation, differentiation, or tissue trafficking. In immunology and infection research, this approach helps identify cell-intrinsic effects of mutations, infections, cytokine signaling, or antimicrobial treatments while controlling for variations in host environment. The results clarify immune-cell fitness and mechanisms that shape protective or pathological responses.

This protocol describes decellularization of Sprague Dawley rat kidneys by antegrade perfusion of detergents through the vasculature, producing acellular renal extracellular matrices that serve as templates for repopulation with human renal epithelial cells. Recellularization and use of the resazurin perfusion assay to monitor growth is performed within specially-designed perfusion bioreactors.
